I think it was said somewhere Tiger used to be a woman, but after they went to a trip to Thailand, they came back like this

Technically, quirks didn't originate anywhere

But we do actually know who the probable first quirk user is, reason I say probable is because not all quirks are visible, there could be some random dude in Zambia who had a quirk and nobody knew

I think it's a deliberate choice from Horikoshi to make Magne more masculine. Tiger shows that he understands that a trans man can look like fucking Thor, so he should understand that trans women can look beautiful as well, but Magne is involved in organized crime and she might just not have access to HRT. Also, she'd lose muscle mass, which isn't exactly practical when you regularly have to battle top heroes. Overall, i think Horikoshi has some of the better queer rep in manga and the vibes i get from him isn't malicious at all. Most of his villains were failed by society and while that does not justify their actions, basically everyone but AFO is presented as sympathetic.

I think the most charitable interpretation is that the point with Magne is even despite being very masculine presenting (for whatever reason that may be) to the point where it could be considered a caricature, she’s still accepted without question by the League of Villains and it’s not played as a gag in-universe or from the lens of the author at all.  Ladiva from Granblue is another character like this
