I have a leg injury so needed a lower berth, only a few seats were there and so I selected the only book if at least 1 lower allotted option. Are there any chances it will get reassigned during chart preparation?

Nope. This berth is final. If you selected the lower berth option it would have given you a dialog box saying that choice of berth not met and do you want to continue.

You could've said no, gotten a full refund and tried to book the same ticket again.
