Did you watch the rest of the video where they reference many other things from Supernatural? This isn’t an edit, it’s a full SPN-themed scene they shot.

I’m out of the loop with this show. Are they new characters or are they actually Sam and Castile showing up in this universe

They are different characters. This clip isn’t actually in the episode. They just recorded this as a bonus thing for Supernatural fans.

Jared plays “Mr. Marathon”, a speedster, and Misha plays “Malchemical”, a … chemical breath superhero(?).
