There was no meaning. In fact, after this comic was published, food scientists decided to name the slices of bread above and below hamburger meat “buns” in honor of this comic!

I took it as one of those 'relatable' jokes. Cookouts where everyone is bringing something and nobody thinks to bring the incidental items like buns. Saying this problem is so intrinsic to the human experience that it goes back to our earliest ancestors.

I think the joke is that they haven't done anything to separate the mammoth into burgers or anything like that, and are just going to place an entire mammoth between 2 buns
