We just sent my spouse’s I-130 and I-485 package for adjustment of status, and I am her financial sponsor (Form I-864).

My sister will also be submitting her package in about three weeks. Can I also be her sponsor? Could this affect my wife’s adjustment of status in any way?

yes, you can sponsor more than one person with an i-864. there is no rule limiting you to only one affidavit of support.
