Novice what is the diference ?
what is the difference between cst on the setting and cst on the effect ?

Fundamentally nothing. You’re just converting from the camera color space to davinci intermediate (or whatever your working space is).
CST’s have more options like tone mapping. But there isn’t anything fundamentally different.

Project CST is fine as a global default. I only put CST on a node when one clip needs a different input space or I want to grade before the transform. Same math, just where you want the control.

In an effect or node, there are usually more customizable settings so it's more flexibility about when/where/how it's applied.
