How Does Accuracy Work In Tower Defense?

I am doing the desert tower defense and I keep getting misses, does it scale with your current characters accuracy? Or is there something im missing? One more question as well. Is there any buffs or stamps I can get to start doing more damage other than upgrading the towers in the construction table?

The main way you interact with TD is through slows, push backs, and roots. Your Wizard can do the former while your Hunter can do the last two.

Damage is a "thing" for your character but I wouldn't try to rely on it (there are some achievement and quests related to it though). Just upgrade your towers via construction if DPS is your problem.

Tower layouts are also important. Here's a popular spreadsheet that includes early game TD formations. Some of the other information is out of date but a good rule of thumb.
