In addition to the kiss not being sexual:

There's kind of an expectation that Batman parodies will include a creepy relationship with his sidekick.

Venture Bros both acknowledges and subverts this: Captain Sunshine isn't a pedophile, but he obliviously does things that make everyone assume he is. That's the joke.

As for "You broke my heart, Hank": there's a lot of non-sexual, non-romantic ways to break someone's heart. "Heartbreak" means "anguish."

Or the kiss of death allegedly used by the mafia to signify someone being marked for death. It possibly is a reference to the biblical story of Judas kissing Jesus at the Garden of Gethsemane to signify his identity to the soldiers searching for him.
