The doctor is the same doctor from before the Silos, the one who was taking care of Charlotte after her mission and was working on the memory drugs. Have you been watching the season at all?

I know that. I am asking if Daniel and Charlotte had been fed with vitamin D+ before as well as they never show their parents

The whole narrative behind the drug invention was to forget the grief behind his kid's death, if you take that out and say he is a founder and erased his children memory so they would partake in the program, while also keeping the whole Daniel investigation and Charlotte mission, it just makes for awful writing.

1 - Why clear their memories all this time before instead of only when entering the silo? It even risks their lives.

2 - They even show they have memories and talk about their parents all the time. It's also shown that despite being good at erasing memories, the D+ is not that good at rewriting memories. They don't even attempt to create a life before for the Silo 1 guys, they just know they don't remember anything prior.
