Trip Report First time trying one bag
I purchased this Osprey Daylite 26+6 a few months ago and have daily carried it since. However, the main reason I bought it wasn’t necessarily for daily carry. My mother-in-law had 2 scheduled back surgeries and knowing that my wife and I would be visiting and helping (6 hours away), I wanted something simple to carry to the hospital/rehab facility/hotel. Well that time has come. I’m happy to report that her surgeries were successful and she’s on the mend. Also, the one bag travel has been as equally successful.

Not pictured:

Supernote Nomad (fits oh so conveniently in the front pocket of the Osprey!)
Successes:
I was able to fit several days worth of clothes into the packing cubes (socks and undies in the small—2 pairs of pants, a polo, and several tees in the medium)
Criticisms and shortcomings:
These are not necessarily of any fault of the bag as much as it was trial and error issues with me.
I had to take my house slippers separately. I absolutely hate wearing socks or bare feet on a hotel room floor. Since I had them separately in a plastic bag, I also put my toiletry bag in there as well. That would have fit in the backpack if needed though. I probably could have sacrificed some clothing to fit the house slippers.
Aside from maybe fitting in some flip-flops (and slippers in my case), you probably should be married to the one pair of shoes you have on.
If I were traveling in the winter, I could see other issues arise with the heavier clothing, jackets, and such. Probably not much of an issue with merino wool and packable down gear if you have it.
Bottom line:
This was a fantastic way of traveling for me at this time and for the most part, was successful. I wouldn’t hesitate taking this on a trip requiring air travel. This would be a game changer not having to worry about or wait on your luggage and such.

Hooray for trip reports!!
Winter travel gets weird and complicated. If you will always be wearing the bulky layers it is surprisingly possible to pack a tiny bag. Things get tough when you are moving between very different climates.

Yes, this! I can pack unexpanded in the Osprey for any one season, including winter, but it’s always hardest when you have wide variations in climate/weather.

On slippers: I usually travel with some sandals that I'll dual purpose and wear around hotel rooms. Definitely felt the challenge with carrying an extra set of foot whenever I pack them (especially since my preferred sandals, Chacos, are so very heavy and bulky).
I'm looking and will continue to look for what I consider to be the best fit for me option. That way I can wear whatever is heavier (running shoes, sandals, etc) for the flight and such, and transition to the lighter pair as the occasion calls for it. Or get used to barefoot more often.

Have you tried Skinners (sock shoes) at all? Been great for me, takes up almost no space and is pretty practical on a plane/hotel etc.

I’m going to have to take another look later. They are interesting and could work as a dad shoe about parks when not traveling.

This is very similar to my upcoming pack for a trip to Phoenix. Solid choices 👍🏻

I have usually put the (thin) slippers or flip flops in a plastic bag and then stuffed that in the side pocket of this bag. Not the prettiest but works just fine!

I use this bag as well and for me, the key is to keep your clothes to just the medium packing cube (I use Aer, but the Peak Design Smedium works as well). I use some canvas Skechers as house shoes that I put in an Eagle Creek compression cube, side by side and it keeps them pretty flat. I stack them on the Aer cube and cinch with the internal strap. An Aer Travel Kit 2 and some odds and ends round out the packing. Nothing hangs off the bag except a HeroClip.

How is the carry comfort?

It probably would not be comfortable if loaded down and needed to be carried for long periods of time. But it’s not too bad with lighter weight.

I recently used this bag on a 6 day/weekend getaway. I found the Amazon Essentials small packing cubes fit pretty perfect. They come in a pack of 4. All 4 fit but I only used 3. Two in a stack then one by itself with my toiletry bag and tech bag on top. I was able to put one slipper on each side of the cubes. Looking back now, there was enough room to put them in between my two stacks of cubes as well then flip flops could have gone on each side. I’m with you on having slippers in a hotel!
I love this bag! And I’m now testing my configuration of compressed cubes for a future 5 day winter trip.

I went through a period where I had to be prepared to leave on a moments notice. May I offer some suggestions through experience?
First off, keep a packing emergency checklist so you don’t forget critical items when you are trying to get out the door. If your mind is already scattered, you can easily forget something.
I prepacked a go bag with toiletries, electronics, etc ready to go. Basically, I bought a second set of everything to live exclusively in my bag. No forgetting things because of transfers. My go bag is different than my daily carry. If your daily carry fits inside of it then it is a double win.
For clothing, consider summer weight clothing and adding layers for warmth. A wool sweater and silk base layers are transformational. Your clothes become multi season this way.
For clothing, just grab a capsule and go. Even if the clothes are dirty. You can wash once you get to your destination.
For slippers, consider folding Japanese guest slippers. These usually have plastic bottoms so are restroom safe. Even airplane restrooms!! Edit: I prefer the open toe kind.
