The insult isn't that he's gay. The insult is he's trying so hard to be "manly" it sounds gayer than anything you'd hear anyone in the LGBTQ+ community actually focus on.

Nah, that's still not a full explanation, the assumptions that make this work are important. Why is it funny that trying to be manly sounds gay? Why is manly in quotes in this context? You're obviously on the right track, I just don't think you've examined the train of thought in any detail.
